The project aims to evaluate whether the Mettl3-mediated m6A modification of Ndrg1 promotes acute kidney injury (AKI) sensitivity in diabetic nephropathy. We will try to answer these questions:
(1) Does upregulation of Mettl3 in DN enhance AKI susceptibility?
(2) Does suppression of Mettl3 in DN attenuate AKI susceptibility?
(3) Is Mettl3-mediated Ndrg1 a major pathway to enhance AKI susceptibility?
(4) Does targeting Ndrg1 knockdown attenuate AKI in diabetic mice?
